Coronavirus is a kind of virus with envelope and linear single strand positive stranded RNA genome. Its diameter is about 80-120nm. At present, it only infects vertebrates such as human, mouse, pig, cat, dog and poultry. New Coronavirus COVID-19 has been identified as a new variant, which can cause viral pneumonia, mild fever, dry cough, heavy breathing difficulties and even shock.
The New Coronavirus (COVID-19) nucleic acid detection kit (QPCR probe method) developed by RXC developed a double target design based on the ORF 1ab (RDRP gene) of New Coronavirus and the specific conserved sequence encoding the N gene of nucleocapsid protein. Using one-step reverse transcription polymerase chain reaction (RT-PCR) combined with TaqMan hydrolysis probe technology, the sample RNA was detected by the change of fluorescence signal on the fluorescence quantitative PCR instrument.
The PCR detection system includes primers and probes for internal standard genes. The process of sample collection and extraction is monitored by detecting whether the internal standard is normal to avoid false negative results.
The New Coronavirus (COVID-19) nucleic acid detection kit (digital PCR method) developed by Rui Xun biology has used the droplet digital PCR technology combined with Taqman fluorescence probe technology to conduct qualitative detection of New Coronavirus.
The samples were randomly assigned to tens of thousands of oil droplets produced by digital PCR chip. The droplet was reacted with PCR. After the PCR reaction was finished, the fluorescence signals of each droplet were read out, and the Poisson distribution principle was used for statistics and analysis, so as to achieve accurate detection of New Coronavirus (COVID-19) specific gene copies.
Among them, covid-19 ORF 1ab detection probe was labeled with FAM fluorescence, N gene detection probe was labeled with hex fluorescence, and internal standard was labeled with Cy5 fluorescence.
